So I want to know if it is possible to do Hunters road from Senyati (Kasane) to Elephant Sands in one day (During August)??

Ask Oom Mac.... I think he has done that road the most recently of all of us..... We drove in April 2014 from Elephants Sands to Padamatenga in a group of 10 VW Kombies.
That was about 1/2 the distance to Kasane.
It was dry.
Mud was no issue. (crossing a few small mud pools ...no issue)
The going was actually "reasonably fast" considering that it was a fairly large group.
One lunch break and two minor breakdown caused by stick penetrating cooling pipes etc.
All in all perhaps 2 hours stationary.
Traveling time from 7:00 am to 4:30pm = 9.5 hours - 2 hours = 7.5 hours driving time.
I would recon that one would reasonably need twice the time to Kasane in dry conditions.
In wet condition ....... different ball-game
